The Seminole Hotel.

Maybe less significant in the grand scheme of things, but I still lament the loss of the Lampru Court.
The tourist would realize at once that he had struck the Land of Flowers - the City Beautiful!

Lampru Court is a poster child for demolition that resulted in blight.
